Tandem scFv

Tandem scFv only consists of the variable regions from distinct antibodies. It is expressed by transfected mammalian cells with four tandem variable regions connected by flexible peptides. Without Fc region, its smaller size (~55 kDa) gives Tandem scFv a short serum half-life, however, it also ensures a closer proximity to the surface of both targets. Among the tandem scFv molecules, there is a special class that can be used to retarget immune cells to disease location. They can be named bispecific immune cell engagers, such as T cell and NK cell. These molecules are able to direct multiple rounds of target cell lysis by immune cells with a low effector cell : target cell ratio. All of these properties have made them one of the most important strategies for cancer treatment and immune regulation.
