Besides the chain paring problem, the affinity and valence of a BsAb to its targets also have an effect on its application potential. Thus, bispecific intrabody is developed to overcome these issues. A bispecific intrabody has two identical immunoglubin chains paired via its recombinant Fc domains. Meanwhile, two pairs of single chain Fvs (scFvs) with distinct specificity are linked to each end of the Fc domain respectively. Since the component unit for dimerization is uniform, this strategy significantly simplifies the generation process of bispecific products. Furthermore, as each component unit is bispecific and bivalence, the homodimer product is not only bispecific but also tetravalence. These properties make bispecific intrabody a leading candidate format for both drug development and academic research.
