Mini-antibody is a group of artificial immunoglobins without CH1 domain. Each single chain of a mini-antibody is expressed separately in prokaryotic or eukaryotic expression system and linked to another signle chain with distinct specificity via chemical modification or knobs-and-holes approach to get bispecific antibody. Removal of CH1 domain does not reduce the affinity of antibody to its targets. On the contrary, the decreased size makes mini-antibodies perform better in tissue penetration than conventional IgG-BsAbs, which is an ideal character for disease treatment and immune research.
